Bacterial contamination can pose a threat to human health and safety. One way to control it in health facilities is to use coatings with antibacterial properties. Copper oxide nanostructures have been proved to possess antibacterial and biocide properties. Laser speckle imaging is an imaging technique based on the analysis of the blurring effect of the speckle pattern. It has been used in numerous studies in wide range of different fields. The aim of our research is to evaluate the application possibilities of this technology in determining the antimicrobial properties of various materials.. A total of 24 different sol-gel CuO surface coatings on polished or phosphatated AISI304 steel (10x3mm) were examined. Coatings contained 0.5,1,3,5 weight % CuO and 1,3 or 5 deposited layers. The antibacterial effect was determined to all samples with novel dried droplet method using E.coli35218. Laser speckle technique was used to determine its potential to be used in evaluation of antibacterial effect, it can visualize submicron movement which can correspond to activity to any object in our case bacteria.. Phosphatized steel coating showed greater antibacterial effect than polished coatings (average antibacterial effect 86.7% vs 35.4%) Using laser speckle technique bacteria activity signal strength were detected. (Values from 0 to 255). To calculate antibacterial effect, control value was subtracted from sample values and those with higher negative value had greater antibacterial effect, 0 or positive value being no antibacterial effect. Almost all phosphatized samples shown antibacterial effect, while only few polished samples had any. Average phosphatized vs polished were -7.6 vs -0.7.. Surface samples deposited on phosphatized steel showed greater antibacterial effect than those deposited on polished steel. Compering both methods, results in most cases were similar, thus showing the potential to use laser speckle method to determine antibacterial effect.
