Detection of SARS-CoV-2 in various clinical specimens using PCR

Description

Purpose: detection of SARS-CoV-2 RNA and load in various clinical specimens of COVID-19 patients. Data include information about 161 COVID-19 patients age, gender, hospitalization status, date of sample collection and presence of SARS-CoV-2 RNA and load in nasopharyngeal swab, plasma, feces, urin and PBMC samples. For 22 out of 161 patients, detection of SRAS-CoV-2 RNA was done in dynamics in one more timepoint ranging from four days to more than two month after first timepoint. RNA was isolated from nasopharyngeal swab, fecal, urine, and plasma samples using Ribospin vRD nucleic acid isolation kit by GeneAll according to the manufacturer’s instructions, and an adapted protocol was used in the case of fecal samples. RNA was isolated from PBMCs with PureZOL by BioRad according to the manufacturer’s instructions. Isolated RNA samples were used to determine the presence of SARS-CoV-2 using Direct SARS-CoV-2 Realtime PCR kit by Vircell and the viral load using quanty COVID-19 kit by Clonit.
